Summary & Explanation
Superoxide dismutase (SOD) plays a critical role in neutralizing harmful free radicals produced during cellular energy generation.  This enzyme accelerates the breakdown, or dismutation, of superoxide radicals into hydrogen peroxide and molecular oxygen.  The activity of SOD can be measured using an indirect assay.  In this assay, xanthine and xanthine oxidase are used to generate superoxide radicals.  These radicals then react with a specific dye, 2-(4-iodophenyl)-3-(4-nitrophenol)-5-phenyltetrazolium chloride, causing it to turn red.  The degree of SOD activity is directly proportional to its ability to inhibit this color change.
