Summary & Explanation
This method builds upon the work of Paglia and Valentine. It utilizes Glutathione Peroxidase (GPx) to catalyze the oxidation of glutathione by cumene hydroperoxide. In the presence of glutathione reductase, the oxidized glutathione (GSSG) is immediately converted back to its reduced form (GSH) while NADPH is simultaneously oxidized to NADP. The activity of GPx is determined by measuring the decrease in absorbance at 340 nm.
